Factors Affecting Glucuronidation in vivo

چکیده
Most foreign compounds are conjugated in vivo producing metabolites which are fairly strong organic acids and are readily excreted in either the urine or bile (see Williams & Millburn, 1974). The major conjugation mechanisms in animals are those utilizing glucuronic acid, glucose, sulphate, glycine, cysteine, acetylation and methylation (see Williams, 1968). Glucuronic acid conjugation is the most widespread and versatile of these conjugation reactions, occurring extensively in man and most mammalian species except the cat. Many foreign compounds with widely different molecular structures can be metabolized by this important conjugation mechanism (see Table l), and possibly this fact together with the ready availability of glucuronic acid from other carbohydrates in the body accounts for the widespread occurrence of glucuronidation. It would not be

Factors affecting glucuronidation activity in Caco-2 cells.
Presystemic intestinal metabolism reduces the intestinal absorption and bioavailability of orally administered drugs. The factors affecting glucuronidation activity in Caco-2 cells seeded in Transwell (4.7 cm(2)) require clarification to establish an in-vitro system to assess intestinal glucuronidation metabolism for novel drug development. alpha-Naphthol (alpha-NA), a substrate for UGT1A6 in C...
متن کاملIn vitro characterization of lamotrigine N2-glucuronidation and the lamotrigine-valproic acid interaction.
Studies were performed to investigate the UDP-glucuronosyltransferase enzyme(s) responsible for the human liver microsomal N2-glucuronidation of the anticonvulsant drug lamotrigine (LTG) and the mechanistic basis for the LTG-valproic acid (VPA) interaction in vivo. LTG N2-glucuronidation by microsomes from five livers exhibited atypical kinetics, best described by a model comprising the express...

The glucuronidation of a number of commonly used hepatic uridine diphosphate glucuronosyltransferase drug substrates has been studied in human tissue microsomes. Prediction of in vivo hepatic drug glucuronidation from liver microsomal data yielded a consistent 10-fold under-prediction. Consideration of protein binding was observed to be pivotal when predicting in vivo glucuronidation for acid s...
متن کاملCarrier-mediated transport of uridine diphosphoglucuronic acid across the endoplasmic reticulum membrane is a prerequisite for UDP-glucuronosyltransferase activity in rat liver.
UDP-glucuronosyltransferases (EC 2.4.1.17) is an isoenzyme family located primarily in the hepatic endoplasmic reticulum (ER) that displays latency of activity both in vitro and in vivo, as assessed respectively in microsomes and in isolated liver. The postulated luminal location of the active site of UDP-glucuronosyltransferases (UGTs) creates a permeability barrier to aglycone and UDP-GlcA ac...
متن کاملInhibitory Effects of Diclofenac on Steroid Glucuronidation In Vivo Do Not Affect Hair-Based Doping Tests for Stanozolol.
In vitro studies show that diclofenac inhibits enzymatic steroid glucuronidation. This study was designed to investigate the influence of diclofenac on the excretion of stanozolol and 3'-hydroxystanozolol via analyses in hair, blood and urine in vivo in a rat study. Brown Norway rats were administered with stanozolol (weeks 1-3) and diclofenac (weeks 1-6). Weekly assessment of steroid levels in...
